We appreciate the reviewer’s constructive suggestion on shake-flask verification after high-throughput preliminary screening. In this study, the oCelloScope 96-well high-throughput static imaging culture was mainly adopted for rapid morphological screening, biofilm formation characterization and strain phenotype primary screening, which realized the rapid elimination of a large number of inferior strains and conditional combinations to save experimental consumption and cycle. After screening out the target dominant strains and optimal medium conditions with significant phenotypic advantages, we have completed the verification test by shaking flask fermentation under conventional aerobic fermentation conditions. The biomass accumulation, metabolic characteristics and functional performance of the target samples in shake-flask culture were consistent with the changing trend of the high-throughput screening results, which verified the reliability of the preliminary screening conclusion. Relevant shake-flask fermentation data have been supplemented in the revised manuscript for detailed display.

本研究依托oCelloScope显微成像平台开展96孔板高通量静态培养初筛,完成多菌株、多培养条件的快速表型初筛与劣样剔除;针对初筛筛选获得的优势菌株与最优培养基配比,统一开展摇瓶振荡发酵验证试验,以摇瓶发酵生物量、代谢产物、菌体形态作为验证指标,印证高通量初筛结果的真实性与适用性,构建“微孔高通量初筛-摇瓶发酵复证”两级试验研究体系。


四、答辩精简口语化解释话术

1. 微孔高通量筛选核心作用是大批量快速初筛,减少低效样品的试验耗材与时间成本,并非直接作为最终定性依据。
